Volgens een nieuwe studie blijkt dat de bioactieve stof apigenine effectief kan zijn bij de bestrijding van kanker. Apigenine is een stof die veel wordt gevonden in bepaalde thee's en groenten en fruit wat gedronken en gegeten wordt in het mediterraandieet, zoals peterselie, selderij, kamille, basilicum enz. Kankercellen hebben hun natuurlijk vermogen om te sterven verloren en blijven daardoor ongebreideld groeien. Uit de studie met borstkankercellen blijkt dat apigenine de kankercellen weer leert om te sterven zodat apoptose of celdood plaats vindt. Scientists Design ‘Fishing’ Technique to Show How Foods Improve Health
COLUMBUS, Ohio – New research suggests that a compound abundant in the Mediterranean diet takes away cancer cells’ “superpower” to escape death.
By altering a very specific step in gene regulation, this compound essentially re-educates cancer cells into normal cells that die as scheduled.
One way that cancer cells thrive is by inhibiting a process that would cause them to die on a regular cycle that is subject to strict programming. This study in cells, led by researchers at The Ohio State University's Comprehensive Cancer Center, found that a compound in certain plant-based foods, called apigenin, could stop breast cancer cells from inhibiting their own death.
Much of what is known about the health benefits of nutrients is based on epidemiological studies that show strong positive relationships between eating specific foods and better health outcomes, especially reduced heart disease. But how the actual molecules within these healthful foods work in the body is still a mystery in many cases, and particularly with foods linked to lower risk for cancer.
Parsley, celery and chamomile tea are the most common sources of apigenin, but it is found in many fruits and vegetables.
The researchers also showed in this work that apigenin binds with an estimated 160 proteins in the human body, suggesting that other nutrients linked to health benefits – called “nutraceuticals” – might have similar far-reaching effects. In contrast, most pharmaceutical drugs target a single molecule.
“We know we need to eat healthfully, but in most cases we don’t know the actual mechanistic reasons for why we need to do that,” said Andrea Doseff, associate professor of internal medicine and molecular genetics at Ohio State and a co-lead author of the study. “We see here that the beneficial effect on health is attributed to this dietary nutrient affecting many proteins. In its relationship with a set of specific proteins, apigenin re-establishes the normal profile in cancer cells. We think this can have great value clinically as a potential cancer-prevention strategy.”

Doseff oversaw this work with co-lead author Erich Grotewold, professor of molecular genetics and director of Ohio State’s Center for Applied Plant Sciences (CAPS). The two collaborate on studying the genomics of apigenin and other flavonoids, a family of plant compounds that are believed to prevent disease.
The research appears this week in the online early edition of the journal Proceedings of the National Academy of Sciences.
Though finding that apigenin can influence cancer cell behavior was an important outcome of the work, Grotewold and Doseff point to their new biomedical research technique as a transformative contribution to nutraceutical research.
They likened the technique to “fishing” for the human proteins in cells that interact with small molecules available in the diet.
“You can imagine all the potentially affected proteins as tiny fishes in a big bowl. We introduce this molecule to the bowl and effectively lure only the truly affected proteins based on structural characteristics that form an attraction,” Doseff said. “We know this is a real partnership because we can see that the proteins and apigenin bind to each other.”
Through additional experimentation, the team established that apigenin had relationships with proteins that have three specific functions. Among the most important was a protein called hnRNPA2.
This protein influences the activity of messenger RNA, or mRNA, which contains the instructions needed to produce a specific protein. The production of mRNA results from the splicing, or modification, of RNA that occurs as part of gene activation. The nature of the splice ultimately influences which protein instructions the mRNA contains.
Doseff noted that abnormal splicing is the culprit in an estimated 80 percent of all cancers. In cancer cells, two types of splicing occur when only one would take place in a normal cell – a trick on the cancer cells’ part to keep them alive and reproducing.
In this study, the researchers observed that apigenin’s connection to the hnRNPA2 protein restored this single-splice characteristic to breast cancer cells, suggesting that when splicing is normal, cells die in a programmed way, or become more sensitive to chemotherapeutic drugs.
“So by applying this nutrient, we can activate that killing machinery. The nutrient eliminated the splicing form that inhibited cell death,” said Doseff, also an investigator in Ohio State’s Davis Heart and Lung Research Institute. “Thus, this suggests that when we eat healthfully, we are actually promoting more normal splice forms inside the cells in our bodies.”
The beneficial effects of nutraceuticals are not limited to cancer, as the investigators previously showed that apigenin has anti-inflammatory activities.
The scientists noted that with its multiple cellular targets, apigenin potentially offers a variety of additional benefits that may even occur over time. “The nutrient is targeting many players, and by doing that, you get an overall synergy of the effect,” Grotewold explained.
Doseff is leading a study in mice, testing whether food modified to contain proper doses of this nutrient can change splicing forms in the animals’ cells and produce an anti-cancer effect.

Door het veranderen van een zeer specifieke stap in genregulatie, deze verbinding in wezen opnieuw opleidt kankercellen in normale cellen die sterven zoals gepland.
Een manier waarop kankercellen gedijen is door remming van een proces dat zou veroorzaken ze sterven op een regelmatige cyclus die zijn strikte programmering. Deze studie in cellen, geleid door onderzoekers van Comprehensive Cancer Center van de Universiteit van Ohio State, vond dat een verbinding in bepaalde plantaardige voedingsmiddelen, genaamd apigenine, kon stoppen borstkankercellen uit het remmen van hun eigen dood.
Veel van wat bekend is over de gezondheidsvoordelen van voedingsstoffen is gebaseerd op epidemiologische studies die een sterke positieve relatie tussen het eten van bepaalde voedingsmiddelen en betere gezondheidsresultaten, vooral verminderde hart-en vaatziekten te tonen. Maar hoe de werkelijke moleculen binnen deze gezonde voedingsmiddelen werken in het lichaam blijft een mysterie in veel gevallen, en met name levensmiddelen verbonden lager risico op kanker.
Peterselie, selderij en kamille thee zijn de meest voorkomende bronnen van apigenine, maar het wordt gevonden in veel groenten en fruit.
De onderzoekers toonden ook deze werk dat apigenine bindt voor een 160 eiwitten in het menselijk lichaam, wat suggereert dat andere voedingsstoffen verbonden gezondheidsvoordelen - zogenaamde "nutraceuticals" - Vertaald verstrekkende gevolgen kan hebben. Doseff overzag dit werk met co-lead auteur Erich Grotewold, hoogleraar moleculaire genetica en directeur van het Centrum van Ohio staat voor Applied Plant Sciences (CAPS). De twee werken samen aan het bestuderen van de genomica van apigenine en andere flavonoïden, een familie van plantaardige verbindingen die worden verondersteld om ziekte te voorkomen.
Het onderzoek verschijnt deze week in de online vroege editie van het tijdschrift Proceedings van de National Academy of Sciences.
Hoewel het vinden van dat apigenine kan kankercel gedrag te beïnvloeden was een belangrijk resultaat van het werk, Grotewold en Doseff punt om hun nieuwe biomedisch onderzoek techniek als transformatieve bijdrage aan nutraceutische onderzoek.
Ze vergeleken de techniek om "vissen" voor menselijke eiwitten in cellen die interageren met kleine moleculen in de voeding.
"Je kunt je voorstellen alle potentieel betrokken eiwitten als kleine vissen in een grote kom. We introduceren dit molecuul aan de kom en alleen de echt getroffen eiwitten gebaseerd op structurele kenmerken die een attractie vormen effectief te lokken, "zei Doseff. "We weten dat dit een echt partnerschap, omdat we kunnen zien dat de eiwitten en apigenine binden aan elkaar."
Door extra experimenten, het team vastgesteld dat apigenine had verhoudingen met eiwitten die drie specifieke functies hebben. Een van de belangrijkste is een eiwit genaamd hnRNPA2.
Dit eiwit beïnvloedt de activiteit van messenger RNA of mRNA, waarbij de instructies die nodig zijn om een specifiek eiwit bevat. De productie van mRNA gevolg van de splitsing of modificatie van RNA die optreedt als onderdeel van genactivering. De aard van de verbinding beïnvloedt uiteindelijk welk eiwit instructies mRNA.
Doseff opgemerkt dat afwijkende splicing is de boosdoener in naar schatting 80 procent van alle kankers. In kankercellen, twee soorten splicing optreden wanneer alleen zou men plaatsvinden in een normale cel - een truc op een deel van de kankercellen 'hen levend en reproduceren te houden.
In deze studie, de onderzoekers opgemerkt dat verband apigenine aan de hnRNPA2 proteïne herstelde deze single-splice eigenschap om borstkankercellen, suggereren dat wanneer splicing normaal sterven cellen in een geprogrammeerde wijze of gevoeliger chemotherapeutica.
"Dus door het toepassen van deze voedingsstof, kunnen we dat het doden van machines te activeren. De voedingsstof geëlimineerd de splicing vorm die celdood geremd, "zei Doseff, ook een onderzoeker in Ohio State Davis Hart en Long Instituut. "Zo, dit suggereert dat wanneer we eten healthfully, we zijn eigenlijk het bevorderen van meer normale splice vormen in de cellen in ons lichaam."
De gunstige effecten van nutraceuticals zijn niet beperkt tot kanker, de onderzoekers toonden eerder aan dat apigenine ontstekingsremmende activiteiten.
De wetenschappers opgemerkt dat met zijn meerdere cellulaire doelwitten, apigenine biedt potentieel een aantal extra voordelen die zelfs kan optreden in de tijd. "De voedingsstof is gericht op veel spelers, en door dat te doen, krijg je een totale synergie van het effect," Grotewold toegelicht.
Doseff leidt een studie in muizen, testen of voedsel gemodificeerd juiste doses van deze voedingsstof bevatten splicing vorm kan veranderen in cellen van het dier en produceren een anti-kanker effect.
